  Probing supramolecular organisation of heterogeneous materials using NMR spectroscopy: from encapsulated pharmaceuticals to soft matter
Molecular level control of the self-assembly of organic molecules at different length scales is a significant challenge both in industrial and academic research. The strength of intermolecular interactions defines physical properties of such materials, i.e. physical stability, solubility, which in turn may affect bioavailability. NMR spectroscopy has been successful in facilitating structural analysis of organic molecular solids. However, there are still considerable challenges when applying structural and dynamic methods across different length and time scales. Combination of experimental and computational approaches can provide molecular level details of the organisation of heterogeneous organic materials with limited ordering (i.e. systems with order-disorder transitions1, encapsulated pharmaceuticals2, 3, polymer/drug composites and soft organogels4, 5). Directing molecular aggregation of pharmaceuticals towards materials with desirable properties can be achieved by their encapsulation inside nanosize crystallisation chambers based on porous solids. Using high-field 19F MAS solid-state NMR methods we have gained unique insight into self-assembly of flufenamic acid (FFA) within the porous solids at different length scales from 4 to 30 nm and identified molecules in three different environments and mobility regimes from confined crystals to liquid-like species3. Using the example of supramolecular hydrogels based on l-phenylalanine we have developed an NMR-based analytical approach to gain insight into the mechanisms of supramolecular gelation4-6. Such materials have unique properties as they combine the well defined crystalline fibres assembled into a hydrogel with significant contribution of solution like domains. Structure and composition of the gel fibres were probed using solid-state NMR and PXRD experiments. Solution-state NMR studies probed the properties of free gelator molecules in an equilibrium with bound molecules (Figure 1B). The dynamics of exchange at the gel/solution interfaces was studied using HR-MAS and saturation transfer difference (STD) NMR experiments.

  Atomic structure/configuration and physical properties studies of hybrid nanostructures by STEM-EELS

In the last two decades, transmission electron microscopes (TEM) have undergone a large number of improvements allowing ~ 100 meV (even few tens of meV) energy resolutions for a sub-nanometer electron beam. These performances offer new possibilities for probing the optical, dielectric and electronic properties of nanomaterials with unprecedented spatial information, as well as for studying the atomic configuration of nanostructures. I will present a selection of recent works taking advantage of these new capabilities. These works will concern the study of the atomic structure & configuration of nanostructures (including doped carbon nanotubes and bio-nanomaterials), as well as opto-electronic properties studies carried out via electron energy loss spectroscopy (EELS) measurements of different kind of nano-objects (inorganic nanotubes and metallic nanoparticles). These works will illustrate the study of properties with extreme spatial resolution enabled by a Cs probe corrected STEM combined with the use of a monochromator.
